NO synthase is within vascular endothelium, makes NO from arginine -> free diffusion across membrane into smooth muscle of tunica media

This is why they don't actually know what nitro does as it would rely on entering the vascular endothelium which apparently doesn't happen at the arterial side where most of those proteins are, since it works on the venule and Coronary artery beds
