Steps to transform a bacteria (insulin)

  1. Mix the plasmids with a bacteria (usually E.coli), some bacteria will take up normal plasmids while others will take up recombinant plasmids. 
  2. To allow the bacteria to take up the plasmids use heat shock or electroporation (the plasma membrane is more permeable to allow the plasmids in)
  3. Incubate the bacteria on antibiotic agar plates (bacteria with recombinant plasmids will survive due to the resistance gene, while bacteria with normal plasmids will die)
  4. Test for the active reporter gene to indicate the bacteria which have taken up recombinant plasmid 
  5. Culture the bacteria so that the genes are expressed and proteins are produced by the bacteria
  6. These fusion proteins are then harvested and purified 
  7. The insulin polypeptides are removed and then combined together to produce functional insulin.
