Steps to transform a bacteria (insulin)

- Mix the plasmids with a bacteria (usually E.coli), some bacteria will take up normal plasmids while others will take up recombinant plasmids.
- To allow the bacteria to take up the plasmids use heat shock or electroporation (the plasma membrane is more permeable to allow the plasmids in)
- Incubate the bacteria on antibiotic agar plates (bacteria with recombinant plasmids will survive due to the resistance gene, while bacteria with normal plasmids will die)
- Test for the active reporter gene to indicate the bacteria which have taken up recombinant plasmid
- Culture the bacteria so that the genes are expressed and proteins are produced by the bacteria
- These fusion proteins are then harvested and purified
- The insulin polypeptides are removed and then combined together to produce functional insulin.
