During DNA synthesis, the 3′ {{c1::OH}} from the last nucleotide of the growing strand "attacks" the {{c1::5′ PO₄}} group of the incoming dNTP.

This reaction releases a pyrophosphate and forms a covalent phosphodiester bond, which links nucleotides together. Mnemonic: 3' OH you wanna fight?
